At 19 years old, he teamed up with fellow Bronx native Chuck Chillout to work for WRKS 98.7 Kiss-FM in New York.

During the early 1990s, Flex made club appearances at Vito Bruno's nightclubs, which led to Joel Salkowitz, a regional vice president of Hot 97, airing live broadcasts from the clubs where Flex was performing. As Hip Hop started gaining momentum again after its decline in the 80s, radio programming increased its hours due to its success.
In the spring of 1992, Funkmaster Flex started hosting his own specialized rap program on Hot 97, which became the first pop station in New York to feature rap music. Since then, he has been with Hot 97 and currently broadcasts nationally through syndication on weeknights and weekends. He is famous for his trademark bomb drop sound effects over records.

During the mid-1990s, Loud Records signed Flex for a set of mixtapes called 60 Minutes of Funk, all four of which were certified gold by the RIAA in the US.
In 1995, he founded The Flip Squad with seven of New York City's most respected disc jockeys, including Biz Markie, BounceMasta Doo Wop, Big Kap, DJ Enuff, Mister Cee, Frankie Cutlass, DJ Riz, Cipha Sounds, and Mark Ronson. Their eponymous debut LP was released on MCA in late 1998.
Flex also released The Tunnel with Def Jam in 1999, which featured tracks by artists such as Dr. Dre, Jay-Z, Eminem, LL Cool J, DJ Myth, Method Man, DMX, Nas, and Snoop Dogg. Furthermore, he has collaborated with Yvette Michele, Pras, DJ Kool, and Armand Van Helden, among others.
In October 2020, Flex announced his plans for an upcoming album. He confirmed that the album would include a variety of artists, such as Juice Wrld, Kodak Black, Post Malone, Lil Baby, and Pop Smoke, among others. In December, he released the album's lead single, Lurkin, with King Von.
