She was born in 1942 in Sorsogon City, Sorsogon, Philippines, and attended St. Agnes Academy before graduating cum laude from St. Theresa's College, a private Catholic women's college in Manila, Philippines.
After completing her education, Lewis earned a law degree from the University of the Philippines College of Law in 1960 and was admitted to the Philippine Bar in 1968.
In 1968, Nicolas-Lewis met her future husband, Reginald F. Lewis, on a blind date in New York City, and they married the following year in Manila. In 1974, Lewis became the first Asian American to pass the American Bar exam without receiving an education in the United States.

She began her career at the Law Students Civil Right Research Council in New York in 1969 and worked at Manhattan Legal Services from 1970 to 1973. From 1979 to 1990, she worked as an attorney for the Immigration and Naturalization Services.
In December 1987, Reginald Lewis made history when he acquired Beatrice International for $985 million, completing the largest leveraged buyout by an African American at that time.
The acquisition created the largest African-American-owned company in the United States.
In 1994, Loida Nicolas-Lewis assumed the position of Chair and CEO of TLC Beatrice International, a $2 billion multinational food company with operations throughout Europe.
She led the company until 2000, after her husband's untimely death. Under her leadership, the company remained successful and continued to grow.
